    Mobile Map Browsers: Anticipated User Interaction for Data Pre-fetching

    When browsing a graphical display of geospatial data on mobile devices, users typically change the displayed maps by panning, zooming in and out, or rotating the device. Limited storage space on mobile devices and slow wireless communications, however, impede the performance of these operations. To overcome the bottleneck that all map data to be displayed on the mobile device need to be downloaded on demand, this thesis investigates how anticipated user interactions affect intelligent pre-fetching so that an on-demand download session is extended incrementally. User interaction is defined as a set of map operations that each have corresponding effects on the spatial dataset required to generate the display. By anticipating user interaction based on past behavior and intuition on when waiting for data is acceptable, it is possible to device a set of strategies to better prepare the device with data for future use. Users that engage with interactive map displays for a variety of tasks, whether it be navigation, information browsing, or data collection, experience a dynamic display to accomplish their goal. With vehicular navigation, the display might update itself as a result of a GPS data stream reflecting movement through space. This movement is not random, especially as is the case of moving vehicles and, therefore, this thesis suggests that mobile map data could be pre-fetched in order to improve usability. Pre-fetching memory-demanding spatial data can benefit usability in several ways, but in particular it can (1) reduce latency when downloading data over wireless connections and (2) better prepare a device for situations where wireless internet connectivity is weak or intermittent. This thesis investigates mobile map caching and devises an algorithm for pre-fetching data on behalf of the application user. Two primary models are compared: isotropic (direction-independent) and anisotropic (direction-dependent) pre-fetching. A prefetching simulation is parameterized with many trajectories that vary in complexity (a metric of direction change within the trajectory) and it is shown that, although anisotropic pre-fetching typically results in a better pre-fetching accuracy, it is not ideal for all scenarios. This thesis suggests a combination of models to accommodate the significant variation in moving object trajectories. In addition, other methods for pre-fetching spatial data are proposed for future research

    MetaCook: FAIR Vocabularies Cookbook

    One of the prerequisites for FAIR data publication is the use of FAIR vocabularies. Currently, tools for the collaborative composition of such vocabularies are missing. For this reason, a universal manual and software for user-friendly vocabulary assembly is being composed in the HMC-funded MetaCook project. The project includes 4 separate test cases from 4 labs across KIT and Hereon, which will help strengthen the software\u27s universality and applicability to various domains. The components described in MetaCook will be implemented in the form of multiple software tools. The first one, a Python-based web application called VocPopuli, is the entry point for domain experts. The software, whose first version is being developed at the time of writing, enables the collaborative definition, and editing of metadata terms. Additionally, it annotates each term, as well as the entire vocabulary, with the help of the PROV Data Model (PROV-DM) - a schema used to describe the provenance of a given object. Finally, it assigns a unique ID to each term in the vocabulary, as well as a hash-based ID the vocabulary itself. The second software tool will facilitate the transformation of the vocabularies developed with the help of VocPopuli into ontologies. It will handle two distinct use cases – the from-scratch conversion of vocabularies into ontologies, and the augmentation of existing ontologies with the terms from a given thesaurus. Both software tools will be used by two semi-overlapping user groups: domain experts will input, edit, and discuss vocabulary terms in their area of interest, while vocabulary and ontology administrators will oversee the vocabulary creation, and ontology transformation. Both the controlled vocabularies and the corresponding ontologies offer the possibility to enrich data documented in Electronic Laboratory Notebooks (ELNs). As the simplest solution, terms used within the ELN are linked to the IDs of the related vocabulary and ontology for an unambiguous definition. Additionally, an export of the defined schemes can be used to automatically create a structured form in the ELNs for documenting the described processes.     Competing signatures of intersite and interlayer spin transfer in the ultrafast magnetization dynamics

    Optically driven intersite and interlayer spin transfer are individually known as the fastest processes for manipulating the spin order of magnetic materials on the sub 100 fs time scale. However, their competing influence on the ultrafast magnetization dynamics remains unexplored. In our work, we show that optically induced intersite spin transfer (also known as OISTR) dominates the ultrafast magnetization dynamics of ferromagnetic alloys such as Permalloy (Ni80Fe20) only in the absence of interlayer spin transfer into a substrate. Once interlayer spin transfer is possible, the influence of OISTR is significantly reduced and interlayer spin transfer dominates the ultrafast magnetization dynamics. This provides a new approach to control the magnetization dynamics of alloys on extremely short time scales by fine-tuning the interlayer spin transfer

    GluN2A NMDA Receptor Enhancement Improves Brain Oscillations, Synchrony, and Cognitive Functions in Dravet Syndrome and Alzheimer's Disease Models.

    NMDA receptors (NMDARs) play subunit-specific roles in synaptic function and are implicated in neuropsychiatric and neurodegenerative disorders. However, the in vivo consequences and therapeutic potential of pharmacologically enhancing NMDAR function via allosteric modulation are largely unknown. We examine the in vivo effects of GNE-0723, a positive allosteric modulator of GluN2A-subunit-containing NMDARs, on brain network and cognitive functions in mouse models of Dravet syndrome (DS) and Alzheimer's disease (AD). GNE-0723 use dependently potentiates synaptic NMDA receptor currents and reduces brain oscillation power with a predominant effect on low-frequency (12-20 Hz) oscillations. Interestingly, DS and AD mouse models display aberrant low-frequency oscillatory power that is tightly correlated with network hypersynchrony. GNE-0723 treatment reduces aberrant low-frequency oscillations and epileptiform discharges and improves cognitive functions in DS and AD mouse models. GluN2A-subunit-containing NMDAR enhancers may have therapeutic benefits in brain disorders with network hypersynchrony and cognitive impairments

    Open and laparoscopic living donor nephrectomy in Switzerland: a retrospective assessment of clinical outcomes and the motivation to donate

    Background. Laparoscopic living kidney nephrectomy is thought to be associated with reduced morbidity, when compared to open nephrectomy. The purpose of this study was to explore the impact of these techniques on donors' clinical outcomes, satisfaction and motivation to donate. Methods. Clinical outcomes were retrospectively compared in 152 open (n = 71) or laparoscopic (n = 81) donor procedures. Donor satisfaction and motivation were assessed with a self-administered questionnaire. Results. The complication rate was the same with both procedures and the majority of complications were mild. Laparoscopy was significantly less painful and resulted in an insignificantly faster return to active life. More than 80% of the donors volunteered to donate without pressure. Worries about future health status, pain or scars were not important in the decision to donate. Similarly, only 15% considered the surgical procedure as instrumental for their decision. Few donors currently worried about their health with one kidney and more than 95% of the donors in both groups stated that they would give their kidney again. Conclusions. Living donor nephrectomy is safe, regardless of the procedure used. Although the laparoscopic nephrectomy offers clear short-term benefits over the open nephrectomy, donors' satisfaction was excellent with both surgical approaches.     A prospective cohort study assessing clinical referral management & workforce allocation within a UK regional medical genetics service

    Abstract Ensuring patient access to genomic information in the face of increasing demand requires clinicians to develop innovative ways of working. This paper presents the first empirical prospective observational cohort study of UK multi-disciplinary genetic service delivery. It describes and explores collaborative working practices including the utilisation and role of clinical geneticists and non-medical genetic counsellors. Six hundred and fifty new patients referred to a regional genetics service were tracked through 850 clinical contacts until discharge. Referral decisions regarding allocation of lead health professional assigned to the case were monitored, including the use of initial clinical contact guidelines. Significant differences were found in the cases led by genetic counsellors and those led by clinical geneticists. Around a sixth, 16.8% (109/650) of referrals were dealt with by a letter back to the referrer or re-directed to another service provider and 14.8% (80/541) of the remaining patients chose not to schedule an appointment. Of the remaining 461 patients, genetic counsellors were allocated as lead health professional for 46.2% (213/461). A further 61 patients did not attend. Of those who did, 86% (345/400) were discharged after one or two appointments. Genetic counsellors contributed to 95% (784/825) of total patient contacts. They provided 93.7% (395/432) of initial contacts and 26.8% (106/395) of patients were discharged at that point. The information from this study informed a planned service re-design. More research is needed to assess the effectiveness and efficiency of different models of collaborative multi-disciplinary working within genetics services.     ThX - a next-generation probe for the early detection of amyloid aggregates.

    Neurodegenerative diseases such as Alzheimer's and Parkinson's are associated with protein misfolding and aggregation. Recent studies suggest that the small, rare and heterogeneous oligomeric species, formed early on in the aggregation process, may be a source of cytotoxicity. Thioflavin T (ThT) is currently the gold-standard fluorescent probe for the study of amyloid proteins and aggregation processes. However, the poor photophysical and binding properties of ThT impairs the study of oligomers. To overcome this challenge, we have designed Thioflavin X, (ThX), a next-generation fluorescent probe which displays superior properties; including a 5-fold increase in brightness and 7-fold increase in binding affinity to amyloidogenic proteins. As an extrinsic dye, this can be used to study unique structural amyloid features both in bulk and on a single-aggregate level. Furthermore, ThX can be used as a super-resolution imaging probe in single-molecule localisation microscopy. Finally, the improved optical properties (extinction coefficient, quantum yield and brightness) of ThX can be used to monitor structural differences in oligomeric species, not observed via traditional ThT imaging
